The somatic activating janus kinase 2 mutation (JAK2)V617F is detectable in most patients with polycythemia vera (PV). Enzymatic assays indicate that both JAK1 and JAK2 are 100- and 20-fold less sensitive to inhibition by CP- 690,550, respectively, when compared with JAK3. JAK2V617F-bearing cells were almost 10-fold more sensitive to CP-690,550 compared with JAK2WT cells, with IC50s of 0.25 μM and 2.11 μM, respectively.
